Abstract

Five broiler chickens of avarage weight average from 1500 – 1700 g were injected intramuscularly with 1mL of BEF vaccine for 4 successive doses in each hen. Antisera were collected and sensitized with latex beads for detection of BEF virus in field samples. The sensitized BEF antibodies (chicken) with latex were tested against 7 mouse brain BEF antigen obtained from Veterinary Serum and Vaccine Research Institute, Abbassia. The results showed clear flacks.
BEF antigen was prepared from Webster 919 strain of BEFV in laboratory after propagation for 6 passages on Vero cells. Cells were collected and concentrated with polyethylene glycol, then centrifuged at high speed centrifuge (50.000 rpm) for 1.5 hour to obtain semi purified antigen. The sediment were resuspended in PBS and used as antigen for coating the plate for measuring antibodies by using indirect ELISA technique, and other part was sensitized with latex particles for detection BEFV antibodies.
A total of 120 nasal swabs (24 buffaloes and 96 cattle) collected from different Governorates of Egypt were tested by prepared latex. The results showed that all buffaloes nasal swabs (24 samples) were negative for detection of BEF antigen while in case of cattle 12 out of 96 nasal swabs were positive for detection of BEF antigen.
