Abstract
Sildenafil citrate is a white to off-white crystalline powder with a solubility of 3.5 mg/mL in water and a molecular weight of 666.7. (sildenafil citrate) is formulated as blue, film-coated rounded-diamond-shaped tablets equivalent to 25 mg. 50 mg and 100 my of sildenafil for oral administration. In addition to the active ingredient, sildenafil citrate, each tablet contains the following inactive ingredients: microcrystalline cellulose, anhydrous dibasic calcium phosphate, croscarmellose sodium, magnesium separate. hydroxypropy methyleellulose. titanium dioxide,lactose, triaeetin, and FD & C Blue #2 aluminum lake (Pfizer-labs., 2004).
Pharmacokinetics: Sildenafil is rapidly absorbed after oral administration reaching maximum plasma concentration within 30-120 minutes of dosing in fasted subjects. It has a bioavailability of about 40% and fatty food has been shown to reduce the rate of absorption and the peak concentrations achieved, without affecting the extent of absorption. It is therefore advised that sildenafil is taken 30-60 minutes before planned sexual activity (Eardly et al., 1998).
