Abstract This study was carried out on the fish samples of about 112 samples from them we found that, two samples infected with A. hydrophila . The mycological and parasitological examination indicated —ye results in all examined samples. For viral diagnosis, conformation and identification of the virus by using cell line from Carp ovary to study the CPE, specific monoclonal antibody for immunohistochemistry, EM and PCR. Were done. -The isolated virus was used for• experimental infection. Clinical signs, PM lesions, histopathological change and change in blood and serum parameters were also examined. The results of this work indicated that: 1-The mortality pattern of the injected Carp indicated that the mortality rate of the silver carp was higher than that of the Common carp. The mortality numbere were 19/30 and 21/30 in Silver carp injected with 0.5 ml X and 1 ml X i03, respectively, while in common carp it was 11/30 and 16/30 for the viral concentrations, respectively. 2- The clinical signs and PM lesions which appeared on the examin(d fish due to SVCV infection were :- The Clinical signs: revealed the presence of skin darkening, tail and fin rot with the postmortem findings of ascitis, exophthalmia with haemorrhagic spots on tie eye of some fish, generalized congestion of internal organs, distended gall bladder’, haemorrhages of gas bladder and oedematous intestine in most examined fish. 3-The tissue culture cell line results indicated that, presence of the virus in the tissue confirmed by CPE formation. The degree of CPE formation was correlated positively with the degree of virus severity and dilution rate of the virus as the high concentration of the viruses the high amount of CPE formation. The CPE formed within 3 — 4 days in which the cells changed from spindle to round and detached forming plaque like. 4-The histopathological investigations revealed that the livers were suffered from that pychnosis, atrophy of parenchyma hepatic cells, the cells became undifferentiated , and the cytoplasmic eosinophilic granules. Also, there was focal necrosis and infiltration of RBCs forming congestion. While, the spleen showed severe congestion of splenie ellipsoids, lymphoid depletion and melanophores aggregations around the blood capillaries and fragmentation of melanomacrophage centers. The histopathologic features in kidney were dilatation of the blood capillaries, thickening of the walls with alteration of the basal lamina, destruction, fibrosis of the glumerulas was observed. Concerning the histopathological features of the intestine, the infected fish showed mild inflammatory and haemorrhage with oedema of submucosa. In case of the histological features of Muscle layer, at experimentally infected Silver Carp showed the main features were common in hyaline degeneration and cloudy swelling of muscular fibers. |
